Small interfering RNA (siRNA) and RT-PCR were used to detect the samples for determine the species and distribution of tobacco virus diseases in Hunan province, and the main virus infection types and the taxonomic status were clarified. The results showed that there were
Tobacco mosaic virus (TMV),
Cucumber mosaic virus (CMV),
Potato virus Y (PVY),
Tomato mottle mosaic virus (TMV),
Rehmannia mosaic virus (ReMV) and
Pepper mild mottle virus (PMMV). Among them, ToMMV, ReMV and PMMV were tested in the first time in Hunan tobacco. TMV, CMV and PVY were the main viruses, and TMV was the dominant virus species, the mix-infection rate was 71.29%. TMV isolates in Hunan tobacco area formed three relatively independent populations, CMV isolates belonged to subgroup IB of group I, and PVY isolates belong to PVY
NTN-a, PVY
E and PVY
SYR-I. In this study, the species, distribution and infection type of tobacco virus in Hunan tobacco area were found out, and the taxonomic status of TMV, CMV and PVY were clarified, which provided a theoretical basis for the prevention and control of tobacco virus diseases.
