    四川和河南烤烟叶片番茄红素β环化酶基因的表达分析

    Analysis on Expression of Lycopene Cyclase Gene in Sichuan and Henan Flue-cured Tobacco (Nicotiana tabacum L.) Leaves in Budding Stage

    • 摘要: 为了探明四川和河南烤烟烟叶类胡萝卜素差异的分子机理,揭示同一品种烤烟在四川和河南产生不同香气风格的机理,采用半定量RT-PCR 和Northern 杂交方法检测了四川和河南烤烟现蕾期叶片中类胡萝卜素合成关键基因番茄红素β 环化酶基因(Lyc-β)的表达水平,并用高效液相色谱法测定了同期烟叶中类胡萝卜素的含量。结果显示,四川烤烟现蕾期叶片中Lyc-β基因的表达水平显著高于河南的;四川烟叶的叶黄素和β胡萝卜素含量显著高于河南的。

       

      Abstract: Abstract:To study the molecular mechanism of the differences in carotenoids between tobacco leaves in Sichuan and Henan, it is primary to reveal that the same species of tobacco will produce different style-type aroma in Sichuan and Henan. Expression of lycopene cyclase gene (Lyc-β) of tobacco (Nicotiana tabacum L.) plants in budding stage was measured by semi-quantitative RT-PCR and Northern blotting in Sichuan and Henan Provinces. Carotenoids contents of tobacco in budding stage were measured by high performance liquid chromatography (HPLC). Results showed that the expression of Lyc-β in Sichuan leaves was higher than that in Henan and the carotenoids contents in Sichuan leaves were significantly higher than those in Henan.
