Abstract:
Two putative TSWV-resistant genes, named
NtSw-5NC89and
NtSw-5zy100 respectively, were isolated from the TSWV-high sensitive
Nicotiana tabacum L. NC89 and TSWV- resistant
Nicotiana tabacumL.zhongyan 100 using homology-based cloning,. Sequence analysis showed that
NtSw-5NC89 is 3819 bp long and contains an open reading frame which encodes a polypeptide of 1273 amino acids with the theoretical isoelectric point 5.64 and molecular mass 147529.5 Da.
NtSw-5 zy100 is 3165 bp long and contains an open reading frame which encodes a polypeptide of 1054 amino acids with the theoretical isoelectric point6.75 and molecular mass 122032.9 Da. No signal peptide or transmembrane region was detected in NtSw-5
NC89 or NtSw-5
zy100 Both proteins contain a CC-(NB-ARC)-LRR typical structure which is shared by most plant resistance proteins. Phylogenetic tree analysis showed that NtSw-5
NC89 had the highest similarity to the
Solanum tuberosum putative late blight resistance protein homolog R1A-3 and NtSw-5
zy100 had the highest similarity to the
Nicotiana sylvestrisputative late blight resistance protein homolog R1A-3 isoform X2. Results of this study provide theoretical basis for resistance study, prevention and treatment of
Nicotianatabacum L. TSWV.
