      抗青枯病雪茄烟ZX145的SNP指纹图谱构建

      SNP Fingerprint Construction and Bacterial Wilt Resistance Identification of Cigar Tobacco ZX145

      • 摘要: 为筛选具有青枯病抗性的优质雪茄烟材料,并为其后续育种应用提供分子标记,本研究基于包含雪茄烟种质ZX145在内的19份烟草种质GBS(基因分型测序,genotyping-by-sequencing)重测序数据,鉴定到221个ZX145特异SNP位点;筛选特异SNP位点开发KASP分子标记,并利用71份不同类型烟草种质进行检测,获得28对能区分ZX145与其他种质的KASP分子标记;挑选覆盖所有染色体的24个位点构成ZX145的特异性SNP指纹图谱;并在福建、贵州、安徽三地病圃开展青枯病抗性及农艺性状、产量性状综合评价。结果表明,ZX145在病圃中抗性表现为中抗至抗病水平,优于常规对照,且产值效益较好。ZX145可作为雪茄烟抗青枯病育种的优质抗源,所得KASP标记可为其育种应用提供支撑。

         

        Abstract: To identify high-quality cigar tobacco germplasm resistant to bacterial wilt (Ralstonia solanacearum) and provide molecular markers for subsequent breeding, this study utilized GBS (genotyping-by-sequencing) resequencing data from 19 tobacco germplasm resources, including the cigar tobacco line ZX145. The number of identified ZX145-specific SNP loci was 221. The specific SNP loci were screened for the development of KASP (kompetitive allele-specific PCR) molecular markers. Validation using 71 tobacco germplasm resources of diverse types yielded 28 KASP marker pairs capable of distinguishing ZX145 from other germplasm. A specific SNP fingerprint for ZX145 was constructed using 24 loci selected to cover all chromosomes. Field evaluations for bacterial wilt resistance, alongside comprehensive assessments of agronomic and yield traits, were conducted in disease nurseries of Fujian, Guizhou, and Anhui Province. The results showed that ZX145 exhibited moderately resistance-resistance in the disease nurseries, which was superior to the conventional control varieties, and its output value performed better. Consequently, ZX145 represents an elite resistant germplasm resource for breeding bacterial wilt-resistant cigar tobacco cultivars. The developed KASP markers provide robust molecular tools to support its utilization in breeding programs.
