      晾晒烟腺毛分泌物抗病毒活性成分鉴定与评价

      Identification and Evaluation of Antiviral Components in Glandular Trichome Secretions of Sun/Air-cured Tobacco

      • 摘要: 为明确晾晒烟腺毛分泌物对植物病毒病的防治效果,挖掘优异活性成分,采用汁液摩擦法测定28份晾晒烟种质腺毛乙醇提取物抗烟草花叶病毒(TMV)活性;并利用超高效液相色谱(UPLC)和气质联用仪(GC-MS)测定了腺毛分泌物二萜、蔗糖酯(SEs)的组成和含量。进一步利用相关、偏最小二乘及聚类方法分析了腺毛分泌物与枯斑抑制率的关系,结果表明:(1)19份种质腺毛乙醇提取物具有抗烟草花叶病毒的活性,其中Q16(湄潭晒烟)腺毛乙醇提取物对烟草花叶病毒的钝化效果最好,枯斑抑制率为68.87%;(2)顺-冷杉醇分别与α-西柏三烯二醇、β-西柏三烯二醇、SEI、SEII显著或极显著负相关,与SEIII、SEIV、SEV显著或极显著正相关;且顺-冷杉醇、SEIII、SEIV蔗糖酯与枯斑抑制率极显著正相关;(3)晾晒烟腺毛分泌物中抗TMV主要活性成分为顺-冷杉醇、SEIII、SEIV,且顺-冷杉醇(200 μg/mL)和蔗糖酯(300 μg/mL)对TMV的钝化效果最好,枯斑抑制率分别为44.32%和53.19%,显著高于植物源抗病毒农药(0.5%氨基寡糖素水剂)。研究结果可为植物源抗病毒活性成分的开发利用提供物质基础和技术支持。

         

        Abstract: In order to elucidate the control effect of glandular trichome secretions from sun/air-cured tobacco on plant viral diseases and to identify superior active components, this study assessed the anti-TMV activity of ethanol extracts from glandular trichomes of 28 sun/air-cured tobacco germplasm resources using a sap rubbing-inoculation method. In addition, the composition and content of diterpenoids and sucrose esters (SEs) in the glandular trichome secretions were determined by ultra-high performance liquid chromatography (UPLC) and gas chromatography-mass spectrometry (GC-MS). Correlation, partial least squares, and clustering analyses were used to investigate the relationship between the content of glandular trichome secretions and the anti-TMV activity. The results showed that: (1) The ethanolic extracts of glandular trichomes from 19 germplasm samples exhibited antiviral activity against TMV, with Q16 (Meitan sun-cured tobacco) showing the best virucidal effect, inhibiting necrotic spots by 68.87%; (2) There was a significant or extremely significant negative correlation between cis-abienol and α-cembratriene-diol, β-cembratriene-diol, SEI, and SEII. Conversely, cis-abienol showed a significant or extremely significant positive correlation with SEIII, SEIV, and SEV. More importantly, cis-abienol, SEIII, and SEIV exhibited a highly significant positive correlation with the inhibition rate of necrotic spots. (3) Cis-abienol, SEIII, and SEIV were identified as the major active components against TMV in the glandular trichome secretions of sun/air-cured tobacco. Additionally, the validation experiments showed that cis-abienol (200 μg/mL) and SEs (300 μg/mL) exhibited the most antiviral activities, with necrosis inhibition rate of 44.32% and 53.19% respectively. These rates were significantly higer than those achieved by a plant-derived pesticide (0.5% amino-oligosaccharides). The research results provide a material foundation and technical support for the efficient utilization and the development of plant-derived antiviral active components.
