    本氏烟tsRNAs对马铃薯Y病毒侵染的响应

    Altered Expression of Transfer-RNA-derived small RNAs after Potato Virus Y Infection in Nicotiana benthamiana

    • 摘要: 为探究本氏烟响应马铃薯Y病毒(potato virus Y, PVY)侵染的分子机制,利用小RNA测序(Small RNA-seq)和转录组测序(RNA-seq)联合分析方法,研究本氏烟在PVY侵染后的tsRNAs和转录响应差异。结果显示,在PVY侵染后第7天,共鉴定出39种差异表达的tiRNAs(tRNA halves)和119种tRFs(tRNA-related fragments)。通过差异表达的靶基因富集分析,发现tRFs在叶片和茎中的117和602个靶基因主要富集在信号转导和能量代谢等通路中。其中,tDR-1:18-His-GTG-1-M3的表达在PVY侵染后的叶片中下调最为显著。在本氏烟中抑制tDR-1:18-His-GTG-1-M3的表达能显著提高其靶基因yabDRNF170的表达水平。该研究结果可为后续深入研究tsRNAs在PVY与宿主植物互作中发挥的作用以及PVY的防治提供参考。

       

      Abstract: To explore the molecular mechanisms underlying Nicotiana benthamiana responses to Potato Virus Y (PVY) infection, small RNA sequencing (sRNA-seq) and transcriptome sequencing (RNA-seq) were employed to analyze the differences in tsRNAs and transcriptional responses in N. benthamiana following PVY infection. The results showed that 39 differentially expressed tiRNAs (tRNA halves) and 119 tRFs (tRNA-related fragments) were identified at the 7th day post-inoculation. Enrichment analysis of differentially expressed target genes (DETGs) demonstrated that 117 and 602 target genes of tRFs in leaves and stems, respectively, were predominantly enriched in pathways such as signal transduction and energy metabolism. Notably, the expression of tDR-1:18-His-GTG-1-M3 was most significantly downregulated in PVY-infected leaves. Suppressing the expression of tDR-1:18-His-GTG-1-M3 in N. benthamiana markedly increasing the expression levels of its target genes, yabD and RNF170. These findings provide a reference for further in-depth research on the role of tsRNAs in PVY-host plant interactions and the development of PVY control strategies.
