      施氮量对不同时期雪茄烟叶细菌群落结构和多样性的影响

      The Effect of Nitrogen Application Rates on the Structure and Diversity of Bacterial Communities in Cigar Tobacco Leaves at Different Periods

      • 摘要: 为明确施氮量对不同时期福建雪茄烟叶细菌群落结构和多样性的影响,本研究以海研204为材料,采用Illumina高通量测序技术,分析不同施氮处理(N0,不施氮;N1~N4,施纯氮120、180、210、240 kg/hm2)雪茄烟叶在田间阶段成熟期、晾制阶段变褐期和干筋期细菌群落结构、多样性和功能。结果表明,不同时期和处理的优势菌门均为变形菌门和放线菌门,优势菌属均为鞘氨醇单胞菌属、甲基杆菌属、金黄色单胞菌属、假单胞菌属,施氮显著影响了变褐期和干筋期鞘氨醇单胞菌属和假单胞菌属丰度。施氮对细菌α多样性影响较小,但显著影响变褐期和干筋期细菌β多样性。成熟期和变褐期,细菌群落主要与碳水化合物代谢、氨基酸代谢、膜转运、能量代谢等功能相关,干筋期细菌群落主要与细胞生长和死亡、细胞运动等功能相关。

         

        Abstract: To clarify the effects of nitrogen application rates on the structure and diversity of bacterial communities in Fujian cigar tobacco leaves at different periods, this study used Haiyan 204 as the test material. Utilizing Illumina high-throughput sequencing technology, we analyzed the results of bacterial community structure, diversity and predicted functions in leaves subjected to different nitrogen treatments (N0, no nitrogen application; N1-N4, 120, 180, 210, 240 kg/hm2 of nitrogen). Analyses were conducted at three key stages: field maturity, and the browning and stem-dry stages during air-curing. The results showed that Proteobacteria and Actinobacteria were the dominant phyla, while Sphingomonas, Methylobacterium, Aureimonas and Pseudomonas were the dominant genera across all periods and treatments. The nitrogen application significantly affected the abundance of Sphingomonas and Pseudomonas during the browning and stem-dry stage of air-curing. Nitrogen application had a small effect on bacterial α diversity, but significantly affected bacterial β diversity in the browning and stem-dry stage in the air-curing periods. In the mature and browning stage, the bacterial community was mainly associated with carbohydrate metabolism, amino acid metabolism, membrane transport, energy metabolism and other functions, while in the stem-dry stage, it was mainly associated with cell growth and death, and cellular motility and other functions.
